Ficoll solution
Ready to use density gradient medium for isolating of PBMC (peripheral blood mononuclear cells - containing lymphocytes and monocytes) out of fresh sample material. It was desinged to isolate cells with a density of less than 1,077 g/cm³ from human whole blood. Antibiotics-free.
This product is intended for research purposes. The product has a Russian Registration Certificate as a medical product.
- Appearance: clear solution, without opalescence and sediment.
- pH: 7.0 to 7.5
- Osmolarity: 280±20 mOsm/kg
- Buffer capacity: at least 1.0 ml
- Endotoxin content: ≤0.125 EU/ml
PREPARATION INSTRUCTIONS: The medium is ready to use. Manipulate the product in aseptic conditions.
METHOD:
- For lymphocytes isolation whole blood is diluted in a ratio of 1:1 with buffer solution of PBS Ca²⁺, Md²⁺ free (or solution of DPBS Ca²⁺, Md²⁺ free).
- The diluted blood is added to Ficoll solution (at the rate of 1.5 volumes of blood per 1 volume of Ficoll solution)/ Avoid a mixing of the two phases.
- Centrifugate for 30 minutes at 400 g at room temperature (18-20 °C).
- Carefully remove the PBMCs from the density gradient medium: whitish meniscus between a plasma and medium.
- The cells are collected and washed in nutrient media for further cultivation and subsequent use for laboratory purposes.
HOW SUPPLIED: in sterile plastic bottle of 100 or 500 ml. Euroflasks have a wide neck, convenient for work.
SHELF LIFE: 24 months. Shelf life after opening the primary packaging: 30 days.
STORAGE: +4 to +30 °C in a place protected from light. After opening the primary packaging, store at temperature of +2 to +8 °C in a place protected from light.
RECOMMENDED USE:
- Do not use the product after its expiry date.
- Use gloves, mask and clothes adapted to the manipulation of the product to avoid contamination.
- Manipulate the product in aseptic conditions.
- Supplements such as antibiotics or L-Glutamine should be added as sterile supplements to the ready medium.
- Medium should be clear and free of opalescence and sediment.
