IMDM powdered medium with glutamine, without sodium bicarbonate, for 50 L
A dry mixture of inorganic salts, amino acids, vitamins, glucose and phenol red. This modification of DMEM was developed for demanding cell cultures. It contains sodium selenite, increased amounts of amino acids and vitamins, and HEPES (25 mmol). Contains no antibiotics. Requires addition of sodium bicarbonate.
Appearance: A yellow to reddish powder, moderately soluble in water.
Intended use: Culture of lymphocytes, bone marrow cells and hybridomas. The increased nutrient content supports rapidly growing or demanding cultures. L-glutamine is already present, so no additional glutamine is required.
Directions for use
- For sterile work, open the plastic container under aseptic conditions.
- Measure 90% of the required final volume of room-temperature cell culture-grade water. While stirring gently, slowly add the weighed amount of powdered medium. Do not heat. Stir until completely dissolved.
- Add the required amount of sodium bicarbonate.
- After all components have dissolved, measure the pH. Adjust with cell culture-grade 1 N HCl or 1 N NaOH. Once the required pH is reached, bring to the final volume, mix gently and measure the pH of the prepared medium.
- Sterilize by filtration through a 0.2 µm or 0.1 µm filter. Aseptically dispense into sterile, tightly sealed containers.
Storage: Store at +2 to +8°C in a dry place protected from light. Prevent moisture from entering the package. Transport by any covered means of transport.
Note: Filtration increases the pH by 0.1–0.2 units.
