Tyrode acidic solution is intended for perforation of the shiny shell (ZONA pellucida) with auxiliary hatching before implantation of embryos.
Tyrode acidic solution is used in assisted reproductive technologies, intended for use in medical and preventive and research institutions.
Tyrode acidic solution basis is purified water, Earl salts, glucose, polyvinylpyrrolidone.
Tyrode acidic solution can be prepared with or without antibiotics.
Sterilized by filtration through filters with pore size of 0.22 microns.
APPLICATION PROTOCOL:
- Before use, Tyrode acidic solution is heated to temperature of 37 ° C.
- A drop (15-20 ml) of Tyrode acidic solution and a few drops of balanced medium IVF 1 "Universal" are placed in the center of the Petri dish. The drops with the medium are coated with balanced liquid paraffin oil, and placed in atmosphere of 5-6% CO2 at temperature of 37 ° C for at least 30 minutes before use.
- For each embryo, one drop should be allocated for manipulation and four drops for washing from the acidic medium.
- For each embryo, the procedure is carried out individually, and each embryo is washed immediately, transferred through 4 drops with IVF medium 1 "Universal", to remove the acidic medium.
- Fill the pipette for auxiliary hatching with Tyrode acidic solution.
- Embryo is fixed with a holding pipette. A pipette with Tyrode acidic solution is brought to Zona pellucida, it should be located at the 3 o'clock position. The pipette should be directed into the empty space in front of the oocyte or into the extracellular space.
- Carefully release solution in the holding area of the pipette end very close to the Zona pellucida. It should take 2 seconds from the moment the solution is released to initiate the process.
- The acidic solution should be stopped leaching as soon as the Zona pellucida has thinned or softened.
- The small hole can be expanded mechanically by moving the micro needle in the hole made.
SHELF LIFE, STORAGE AND TRANSPORTATION CONDITIONS
Shelf life is 3 months from the date of manufacture, subject to the storage regime.
Store and transport in accordance with SR 3.3.2.1248-03 at temperature of 2 to 8 ° C in a place protected from light.
Do not freeze!
Unused residue of the heated medium is not subject to reheating and use.
The product must be used within 7 days from the bottle first opening.
It is recommended to store the medium with a minimum volume of air in the bottle.
Open the vial in sterile conditions.
