HybriS-1-CHO medium without glutamine, 500 mL, EuroPET
A serum-free medium for suspension culture of CHO cells, including recombinant clones and cell lines. Consists of inorganic salts, amino acids, vitamins, glucose and synthetic serum substitutes dissolved in deionized water and sterilized by filtration through a 0.22 µm filter. Contains no hydrolysis products of animal or plant proteins, glutamine sources or nucleotides. Contains insulin.
Product: A clear, yellowish liquid without opalescence or precipitate. Specific activity in a bioreactor suspension culture of CHO-S cells, seeded at 5×10⁵ cells/ml and assessed on day 4: doubling time 15.0 hours, specification not more than 26 hours. Contains no antibiotics or glutamine.
Intended use: Intended for establishing and efficiently maintaining suspension cultures of recombinant CHO clones and cell lines in biotechnology processes. Enables rapid adaptation of adherent CHO cultures grown with serum to serum-free suspension conditions. Maximum viable CHO-S cell density reaches 0.8–1.6×10⁷ cells/ml on culture days 3–5.
Directions for use:
- Open the bottle under aseptic conditions;
- Add the selective supplements and nutrients needed for growth and recombinant product expression, depending on the specific CHO line;
- For clones deficient in dihydrofolate reductase (DHFR) or glutamine synthetase (GS), supplement the medium with nucleotide precursors in the form of hypoxanthine/thymidine (HT), or with glutamine (alanyl-glutamine);
- Dispense the prepared medium into culture vessels or bioreactor culture systems;
- Passage cells at a final density of 1.0–10.0×10⁵ cells/ml. Culture at 37°C, 5–6% CO₂ and 115–135 rpm;
- Specialized feeds, Feed No. 1 and Feed No. 5, which contains insulin, are recommended to achieve high cell densities and productivity. Add glutamine according to the formulation before use.
Storage conditions: Store and transport in the manufacturer's packaging at 2–8°C, away from heat and light. Shelf life after opening the primary packaging: 1 month from the date the bottle is opened. Transport by any covered means of transport under the specified conditions.
