IVF medium"Droblenie" is intended for cultivation of embryos with a change of medium. In IVF medium "Droblenie" embryos develop within 48 hours (day 2 after ICSI or fertilization). Then there is a change of medium for IVF PRO "Blastnaya".
IVF medium"Droblenie" is used in assisted reproductive technologies, is intended for use in medical and preventive and research institutions.
The main characteristics:
IVF medium"Droblenie" is prepared on the bicarbonate buffer basis.
Composition: purified water, mineral salts, carbohydrates, amino acids, vitamins, human serum albumin. Albumin was tested for presence of hepatitis B surface antigen, for antibodies to hepatitis C viruses, HIV-1 HIV-2, for antibodies to the causative agent of syphilis.
IVF medium"Droblenie"can be prepared either with the addition of antibiotics and phenol red, or without these components (at customer’s request ).
IVF medium"Droblenie"is aseptically prepared by sterilizing filtration.
The bottle opening and further work with the medium should be carried out in sterile conditions!
Application Protocol
1. Before use, the slightly opened bottle with the medium is kept in atmosphere of 5-6% CO2 at 37 °C for 2 hours.
2. During the ICSI procedure (Day 1), oocytes, after sperm injection, are kept in a fertilization medium (as such, it is recommended to use IVF 1 "Universal" medium), until two pronuclei are formed (after 16-18 hours). And then, the fertilized oocytes with two pronuclei are moved to an IVF medium «Droblenie », aged in a CO2 incubator, where further cultivation takes place.
3. After fertilization (Day 1), successive movements of zygotes are carried out through drops with balanced IVF medium "Droblenie" to wash off the cells of the cumulus complex and spermatozoids.
4. After washing, the formation of pronuclei is checked, and zygotes with two pronuclei are used for further cultivation.
5. Cultivation can be carried out in microdrops with medium coated with liquid paraffin oil, or in 500 µL of medium without oil coating.
6. To continue cultivation, embryos at the 4-8 cell stage are carefully washed with balanced IVF PRO "Blastnaya" medium and transferred to a new portion of the same medium, previously balanced in atmosphere of 5-6% CO2 at temperature of 37 °C.
7. If it is necessary to transfer embryos into the uterus on day 4 or 5 of cultivation, medium for transferring IVF PRO "Shvartovaya" in a volume of 20-30 µl is used, after balancing it in atmosphere of 5-6% CO2 at temperature of 37 °C for 2 hours.
Shelf life -3 months from the date of manufacture, subject to the storage regime.
Store and transport the medium at temperature of 2 to 8 °C in a place protected from light.
Do not freeze!
Unused residue of the heated medium is not subject to reheating and use.
The product must be used within 7 days from the first bottle opening.
It is recommended to store the medium with a minimum volume of air in the bottle.
