Perselect medium, 100%, 10 mL
Perselect medium is manufactured to specification TU 21.20.23-015-45171785-2017.
A ready-to-use mixture of silane-coated colloidal silica particles based on Percoll PLUS, containing salts, amino acids, carbohydrates and the antibiotic gentamicin sulfate. Sterilized by membrane filtration.
Appearance: A translucent opalescent liquid without precipitate.
Intended use: Isolation of sperm from semen by density gradient fractionation in preparation for assisted reproductive technology procedures. It can be used to prepare gradients from 35 to 90% in IVF clinics and reproductive medicine centres.
Directions for use
Perselect 100% contains gentamicin sulfate. Healthcare professionals with a known allergy to gentamicin sulfate or related antibiotics should avoid direct contact with the product and follow the instructions for use.
Gradient preparation: Layer 1 mL of Perselect 45% over 1 mL of Perselect 90%.
Semen preparation: Incubate the collected semen for 20 minutes at room temperature, then mix thoroughly. If the sample is too viscous, it may be passed through a narrow-bore pipette. Assess sperm concentration and motility microscopically.
Sperm suspension preparation:
- Carefully layer 1 mL of semen onto the prepared gradient. Do not increase the semen volume to more than twice the gradient volume, as this may impair separation.
- Centrifuge the gradient at 400 g (1500 rpm) for 20 minutes at room temperature.
- Remove the supernatant, resuspend the pellet in 5 mL of IVF 1 Spermoprep or IVF 1 Universal medium, and centrifuge at 1500 rpm for 10 minutes.
- Repeat the washing procedure.
- Remove the supernatant and resuspend the pellet in an appropriate volume of IVF 1 Spermoprep or IVF 1 Universal medium.
Use the medium within 1 month of first opening the bottle. Do not reheat or reuse any unused portion of warmed medium.
Storage: Store at +2 to +8°C, protected from light. Transport by any covered means of transport. Do not use medium transported outside the specified temperature range. Do not freeze.
