« Perselect » media 100%
Ready-to-use media. It is a mixture of colloidal silicon particles with a silane coating based on Percoll PLUS. It contains salts, amino acids, carbohydrates, antibiotic-gentamicin sulfate.
The media provide natural environmental conditions to isolation of spermatozoid from the ejaculate by gradient fractioning in preparation for assisted reproductive technologies.
The "Perselect" media are 100% made for preparation of the required density gradients. If necessary, it is possible to prepare gradients with a density in the range of 35-90%.
This product has a Registration Certificate No. RZN 2019/5717 dated November 21, 2019.
The “Perselect” media are 100% used in assisted reproductive technologies, as well as in centers of reproductive medicine and in vitro fertilization (IVF).
The media are made under aseptic conditions by sterilizing filtration through filters with pore size of 0.22 microns. It is translucent opalescent liquid without sediment. It is available in polyethylene terephthalate hermetically sealed bottles with single-component polyethylene caps with the first opening control.
The bottle opening and further work with "Perselect" media should be in sterile conditions.

Gradient preparing
To prepare the gradient, 1 ml of the "Perselect" media 45% is layered on 1 ml of the “Perselect" media 90%.
Ejaculate preparation
The collected ejaculate is incubated for 20 minutes at room temperature, then thoroughly mixed. If the sample is too viscous, it is allowed to pass it through a pipette with a narrow tip.
The concentration and motility of spermatozoids are evaluated microscopically.
Preparation of spermatozoid suspension
- * Carefully layer 1 ml of ejaculate on the surface of the prepared gradient. It is not recommended to increase the amount of ejaculate in the ratio to the gradient by more than 2 times, as this can lead to poor-quality separation.
- Gradient is centrifuged at acceleration of 400g (1500 rpm) for 20 minutes at room temperature.
- * Supernatant is removed, the precipitate is resuspended in 5 ml of IVF 1 "Spermoprep" or in IVF 1 "Universal" and centrifuged at 1500 rpm for 10 minutes.
- Washing procedure is repeated
- * Supernatant is removed, the precipitate is resuspended in the appropriate extent of IVF 1 "Spermoprep" media or in IVF 1 "Universal".
Precautions
"Perselect" media 100% contain antibiotic gentamicin sulfate. Medical workers with detected allergy to gentamicin sulfate or similar antibiotics who start working with the media should avoid direct contact with the product and act according to the Instructions for using the “Perselect” media 100%.
Shelf life, storage and transportation conditions:
Shelf life is 12 months from the date of manufacture.
The media should be used within 1 month from the moment of the bottle first opening.
Do not reheat and do not use media remainder.
Store and transport at temperature of +2 to +8 °C in a place protected from light.
Do not use "Perselect" media 100%, transported with temperature regime violation.
Do not freeze!
