"Perselect" media 90%
Ready-to-use media. It is a mixture of colloidal silicon particles with silane coating based on Percoll PLUS. It contains salts, amino acids, carbohydrates, antibiotic - gentamicin sulfate, human albumin.
Provide natural environmental conditions to sperm isolation from the ejaculate by gradient fractioning in preparation for procedures of assisted reproductive technologies.
« Perselect » media 90% are intended for preparation of single-stage gradient.
"Perselect" media 90% can be used both individually (single-stage gradient) and in combination with "Perselect" media 45% (two-stage gradient), depending on floating density and the required purification degree of the selected cells fractioning.
For the best results, the gradient is prepared immediately before use.

« Perselect » media are 90% used in assisted reproductive technologies, as well as in centers of reproductive medicine and in vitro fertilization (IVF).
« Perselect » media contain 90% human serum albumin tested for antigen to the hepatitis HBsAg virus - free and for absence of antibodies to HIV-1/-2, HIV-1, hepatitis B and C viruses and parvovirus B19.
The media are produced under aseptic conditions by sterilizing filtration through filters with pore size of 0.22 microns. It is light pink opalescent sediment-free liquid. It is available in hermetically sealed polyethylene terephthalate bottles with single-component polyethylene caps with first opening control.
Open the vial and work with « Perselect »media in sterile conditions.

Gradient preparation
To prepare two-stage gradient, 1 ml of "Perselect" media 45% is layered on 1 ml "Perselect" media 90%.
Preparation of ejaculate
The collected ejaculate is incubated for 20 minutes at room temperature, then thoroughly mixed. If the sample is too viscous, it is allowed to pass through a narrow tip pipette.
Concentration and motility of spermatozoids are evaluated microscopically.
Preparation of sperm suspension
Carefully layer 1 ml of ejaculate on the prepared gradient surface. It is not recommended to increase the amount of ejaculate in relation to the gradient by more than 2 times, as this can lead to poor-quality separation.
The gradient is centrifuged at acceleration of 400g (1500 rpm) for 20 minutes at room temperature.
Supernatant is removed, precipitate is resuspended in 5 ml of IVF 1 "Spermoprep" or in IVF 1 "Universal" and centrifuged at 1500 rpm for 10 minutes.
The washing procedure is repeated.
The supernatant is removed, the precipitate is resuspended in the appropriate volume of IVF 1 "Spermoprep" media or in IVF 1 "Universal".
Contraindications of medical device
« Perselect » media contain 90% gentamicin sulfate antibiotic. Medical workers with detected allergy to gentamicin sulfate or similar antibiotics, starting to work with the media, should avoid direct contact with the product and act according to the Instructions for use of the "Perselect"90% media.
Shelf life, storage and transportation conditions
Shelf life is 6 months from the production date.
The media should be used within 7 days from the first bottle opening.
The unused heated media residue are not subject to reheating and use.
Store and transport at temperature of +2 to +8 ° C in a place protected from light.
The "Perselect" 90% media, transported with violation of the temperature regime, are not subject to use.
