Choriokar-1 test kit for cell karyotyping, Romanowsky staining
A ready-to-use reagent kit for culturing chorionic villus cells and preparing fetal metaphase chromosomes for karyotyping. A distinguishing feature of the method is cell maceration in acetic acid after standard fixation.
Kit contents:
- Test bottles with medium for villus aspiration and transport: 2 bottles of 30 mL, sterile;
- Test bottles with medium for villus washing: 2 bottles of 8 mL, sterile;
- Test bottles with medium for cell incubation: 2 bottles of 5 mL, sterile;
- Colchicine: 1 bottle, 0.5 mg;
- Sodium citrate: 1 bottle, 100 mg;
- Romanowsky azure-eosin stain: 1 bottle, 10 mL;
- Pipettes: 2 pcs., 3 mL each.
Intended use: Used to incubate chorionic villus cells for direct or semi-direct preparations from sporadically dividing cells and subsequent analysis of fetal metaphase (mitotic) chromosomes. Colchicine arrests mitosis; sodium citrate solution is used for hypotonic treatment; Romanowsky azure-eosin stain is used for staining.
Directions for use: Place the chorionic villus sample in 15–20 mL of transport medium, wash, and transfer to incubation medium. Incubate at 37°C for 18–20 hours. Add 30 µl of colchicine solution per bottle, prepared by dissolving 0.5 mg in 5 mL water, and incubate for 2 hours. Perform hypotonic treatment with 1% sodium citrate for 30 minutes at 37°C, followed by fixation in methanol–acetic acid, 3:1. Macerate cells in 60% acetic acid on a microscope slide at 56–60°C. Stain with Romanowsky azure-eosin using the standard method. For sterile work, open bottles under aseptic conditions.
Storage conditions: Store at −10 to −20°C, protected from light.
